Roll 3d6 six times.
If you roll 5 or less, drop that entire ability and replace it with another that that you make up. Drop only one ability per character. Imagine part of a whole system with this new altered ability set.
Example:
STR 6 INT 10 WIS 7 CON 5 DEX 12 CHA 10
Drop CON, replace it with BLG (Body Language). Represents ability to communicate subtle cues with one's face and body. Typical checks represent reality, that is, result can be positive or negative depending on character's desire. Intimidation might use this score, but a high score might also give you away when trying to bluff.
All scores may be re-interpreted to have possibe negative effects as well, with possible mechnical consequences.

But what do you do if you need their constitution score? Just an exercise by which to generate a new ability system. Not a way to run a character in a D&D-like system that requires all six of the usual scores, of course!

Actually, it's an interesting idea... yes, you have a horrible constitution, but you have a great score in this random ability that not everyone has! Maybe make the other ability 21-<poor ability score> to give the same distance from the extreme; sure, you have a 5 in CON, but now you have a 16 in BLG with the attendant bonuses.

The idea was SUPPOSED to be 1) Roll as if rolling up ability scores. 2) If one is low, replace it with something you just made up. 3) Then make a game system, or the seed for one, based on those abilities. Not "give one character this random score." But I like what you did above, too.
